The blaze, reported around 9 am, triggered a massive rescue operation involving multiple firefighting vehicles.
More than 40 people were rescued and shifted to nearby hospitals, according to local media reports.
Visuals from the scene showed people climbing down ladders and jumping from the building to escape the flames.
The majority of those killed and critically injured are foreign nationals. Delhi Police have initiated legal action against the hotel owner.
They are conducting raids to arrest him, aiming to invoke culpable homicide charges.
Investigators are examining possible fire safety violations, with preliminary findings indicating that the B&B did not possess a mandatory Fire No Objection Certificate (NOC), the India Today reported on Wednesday.
Prime Minister Narendra Modi and the Delhi government expressed condolences and announced assistance for victims.
He assured support to affected families.
